The inhibitory effects of PEG on whole-plant growth can exceed the effects of other osmolytes such as NaCI, and this has been ascribed to toxic contaminants, or to reduced oxygen availability in PEG solutions. We investigated another possibility, namely that PEG has an additional inhibitory effect on root water transport which in turn affects leaf development. The effects on first-leaf growth of applications of PEG 6000 or isoosmotic NaCI to the roots were determined using hydroponically grown maize (Zea mays L.) seedlings. Leaf growth rates were inhibited within minutes of PEG application to the roots and remained inhibited for days. The inhibitory effects on growth of NaCI, and also of KCl and mannitol, were much smaller. The comparative effects of NaCI and PEG on root water transport were determined by assaying pressurized flow through excised roots. PEG induced a 7-fold greater inhibition of flow through live roots than NaCI. Killing of the roots by heat treatment, to reduce cell membrane resistances to solute penetration, nearly doubled the flow rate for roots in NaCI, but not for roots in PEG. We suggest that the greater viscosity of PEG solutions, as compared with NaCI, may be a primary factor contributing to the additional inhibition of water flow through live and killed roots. PEG did not have additional effects on leaf turgor but had a 3 times greater inhibitory effect than NaCI on the irreversible extensibility of the leaves and induced 16 times more leaf accumulation of the growth inhibitory stress hormone abscisic acid (ABA). We conclude that greater inhibition of root water transport by PEG 6000, as compared with NaCI, leads to additional reductions in extensibility, additional ABA accumulation, and a greater inhibition of leaf growth. 相似文献

The 5? cap and 3? poly(A) tail of mRNA are known to synergistically stimulate translation initiation via the formation of the cap?eIF4E?eIF4G?PABP?poly(A) complex. Most mRNA sequences have an intrinsic propensity to fold into extensive intramolecular secondary structures that result in short end-to-end distances. The inherent compactness of mRNAs might stabilize the cap?eIF4E?eIF4G?PABP?poly(A) complex and enhance cap-poly(A) translational synergy. Here, we test this hypothesis by introducing intrinsically unstructured sequences into the 5? or 3? UTRs of model mRNAs. We found that the introduction of unstructured sequences into the 3? UTR, but not the 5? UTR, decreases mRNA translation in cell-free wheat germ and yeast extracts without affecting mRNA stability. The observed reduction in protein synthesis results from the diminished ability of the poly(A) tail to stimulate translation. These results suggest that base pair formation by the 3? UTR enhances the cap-poly(A) synergy in translation initiation. 相似文献

根系构型决定了植物对资源的吸收方式,根系构型的变化是植物对环境的生态适应和有效生存策略。在阿拉善高原西南缘红砂(Reaumuria soongarica)-珍珠猪毛菜(Salsola passerina)混生群落采用传统挖掘法收集两种植物根系,基于量化的根系形态指标,利用几何拓扑学及分形理论分析了根系构型特征,探讨了该地区2种植物对干旱生境的生态适应策略。结果表明:红砂和珍珠猪毛菜根系均以水平分布占优,根系浅层化分布明显,混生的两种植物占据不同的生态位;2种荒漠植物均具有较大的比根长(SRL)和比表面积(SRA),红砂SRL=21.3 cm/g,SRA=7.6 cm2/g,珍珠SRL=22.4 cm/g,SRA=6.5 cm2/g,有利于水分和养分的获取;红砂根系拓扑指数(TI)、修正拓扑参数(qa和qb)分别为0.86、0.52、0.49,珍珠猪毛菜对应参数分别为0.93、0.76、0.73,表明2种植物根系均趋向于鱼尾形分支结构;根系分形维数值(FD红=1.488、FD珍=1.422)较小,而分形丰度值(lgK红=1.855、lgK珍=1.774)较大,表明2种植物分支相对简单,但空间拓展能力强,有利于对营养空间的占有。上述特征可能是阿拉善西南缘红砂-珍珠猪毛菜群落2种荒漠植物植物对干旱贫瘠生境的重要生态适应策略。 相似文献

Data are presented suggesting that birds have evolved eggs with shells containing different structures (numbers of mammillae per unit of inner eggshell surface area, i.e., mammillary densities) to cope up with different calcium requirements imposed by different growth rates and modes of development. Precocial bird species grow slowly, but have high mammillary density, while altricial bird species grow rapidly, but have low mammillary density. These results suggest an adaptation associated with growth rate and mode of development and show, moreover, that the mammillary layer is indicative of the breeding biology of the bird. J. The capacity of preformation and neoformation and the structure of winter buds are vegetative attributes that may vary between plant species and according to ontogenetic stages of the same species. The present study describes and evaluates these features for the four tree species of Proteaceae occurring in Patagonia. In particular, it analyzes the structure and development of the distal buds of the trunk and the preformed or neoformed nature of the organs involved. Two of the species, Embothrium coccineum and Lomatia hirsuta, have scaly buds, in which primordia of green leaves are covered by cataphylls. The shoots of both species may include neoformed organs, more frequently so in juvenile trees. Lomatia ferruginea and Gevuina avellana have naked buds with a low number of primordia; in juvenile and adult trees of both species trunk shoots are entirely preformed. The structure of buds and shoots suggests two different growth modalities of the axes, which would be related to ecological breadth (narrower in the species with naked buds than in those with scaly buds) and distribution area of these species. The considerable morphological differences between the two Lomatia species studied raise the question whether they ought to be included in the same genus. 相似文献

The human endolymphatic duct (ED) and sac of the inner ear have been suggested to control endolymph volume and pressure. However, the physiological mechanisms for these processes remain obscure. We investigated the organization of the periductal interstitial connective tissue cells and extracellular matrix (ECM) in four freshly fixed human EDs by transmission electron microscopy and by immunohistochemistry. The unique surgical material allowed a greatly improved structural and epitopic preservation of tissue. Periductal connective tissue cells formed frequent intercellular contacts and focally occurring electron-dense contacts to ECM structures, creating a complex tissue network. The connective tissue cells also formed contacts with the basal lamina of the ED epithelium and the bone matrix, connecting the ED with the surrounding bone of the vestibular aqueduct. The interstitial connective tissue cells were non-endothelial and non-smooth muscle fibroblastoid cells. We suggest that the ED tissue network forms a functional mechanical entity that takes part in the control of inner ear fluid pressure and endolymph resorption. 相似文献

Surface-coat epitopes of Meloidogyne incognita were detected in root tissues of Arabidopsis thaliana during migration and feeding site formation. A whole-mount root technique was used for immunolocalization of surface coat epitopes in A. thaliana, with the aid of a monoclonal antibody raised specifically against the outer surface of infective juveniles of M. incognita. The antibody, which was Meloidogyne-specific, recognized a fucosyl-bearing glycoprotein in the surface coat. During migration in host tissues the surface coat was shed, initially accumulating in the intercellular spaces next to the juvenile and later at cell junctions farther from the nematode. A large number of trafficking steps occur between the last compartment of the Golgi apparatus (TGN) and the vacuole of the yeast Saccharomyces cerevisiae. To date, two intracellular routes from the TGN to the vacuole have been identified. Carboxypeptidase Y (CPY) travels through a prevacuolar/endosomal compartment (PVC), and subsequently on to the vacuole, while alkaline phosphatase (ALP) bypasses this compartment to reach the same organelle. Proteins resident to the TGN achieve their localization despite a continuous flux of traffic by continually being retrieved from the distal PVC by virtue of an aromatic amino acid–containing sorting motif. In this study we report that a hybrid protein based on ALP and containing this retrieval motif reaches the PVC not by following the CPY sorting pathway, but instead by signal-dependent retrograde transport from the vacuole, an organelle previously thought of as a terminal compartment. In addition, we show that a mutation in VAC7, a gene previously identified as being required for vacuolar inheritance, blocks this trafficking step. Finally we show that Vti1p, a v-SNARE required for the delivery of both CPY and ALP to the vacuole, uses retrograde transport out of the vacuole as part of its normal cellular itinerary. 相似文献

Bile micelles play an important role in oral absorption of low‐solubility compounds. Bile micelles can affect solubility, dissolution rate, and permeability. For the pH–solubility profile in bile micelles, the Henderson–Hasselbalch equation should be modified to take bile‐micelle partition into account. For the dissolution rate, in the Nernst–Brunner equation, the effective diffusion coefficient in bile‐micelle media should be used instead of the monomer diffusion coefficient. The diffusion coefficient of bile micelles is 8‐ to 18‐fold smaller than that of monomer molecules. For permeability, the effective diffusion coefficient in the unstirred water layer adjacent to the epithelial membrane, and the free fraction at the epithelial membrane surface should be taken into account. The importance of these aspects is demonstrated here using several in vivo and clinical oral‐absorption data of low‐solubility model compounds. Using the theoretical equations, the food effect on oral absorption is further discussed. 相似文献

The circadian clock regulates a wide range of electrophysiological and developmental processes in plants. Here, we discuss the direct influence of a circadian clock on biologically closed electrochemical circuits in vivo. The biologically closed electrochemical circuits in the leaves of C. miniata (Kaffir lily), Aloe vera and Mimosa pudica, which regulate their physiology, were analyzed using the charge stimulation method. Plants are able to memorize daytime and nighttime. Even at continuous light or darkness, plants recognize nighttime or daytime and change the input resistance. The circadian clock can be maintained endogenously and has electrochemical oscillators, which can activate ion channels in biologically closed electrochemical circuits. The activation of voltage gated channels depends on the applied voltage, electrical charge, and the speed of transmission of electrical energy from the electrostimulator to plants. 相似文献
